Benjamin Franklin Gates has dedicated his life to guarding a treasure that most people think doesn't exist. A responsibility handed down over four generations, Gates has spent his life arguing with his non-believing father about the secret Knights of the Templar treasure, a legendary hoard that brings not only riches but power and control.
The treasure has been held by the most powerful nations on earth since the Pyramids were constructed in Egypt. Gates' problem is that even he doesn't know where the treasure is, until he becomes the first person in 223 years to unlock the clues that are all around us. The key to the treasure is a map that has been hidden in plain sight since the American Revolution, written on the back of the Declaration of Independence, which is held safe in the National Archives behind the greatest security system yet invented. But that won't stop Ian Howe, Gates's greedy and ruthless adversary who has no understanding of the treasure's power.
Gates is forced to sacrifice everything for his country and steal the Declaration in order to save it. Teaming up with Dr. Abigail Chase (DIANE KRUGER), the beautiful, brilliant director of the Archives, Gates aims to stay one step ahead of Howe in a tense, exciting battle of wits.
太古の昔より、その《秘宝》は常に《至高の権力》と共に存在した－。紀元前2000年、エジプトのファラオの墓に、エルサレムのソロモンの神殿に、フランスのテンプル騎士団のアジトに、コロンブスを乗せたサンタマリア号に、アステカの太陽のピラミッドに・・・・その秘宝の近くには、いつも“全能の眼と未完成のピラミッド”からなるマークが記されていた。いくつもの大陸をめぐった秘宝は1779年、独立戦争が激化するアメリカで忽然と姿を消す。そして－。
2004年－歴史学者にして冒険家のベンジャミン（ベン）・ゲイツ（ニコラス・ケイジ）は、一族の夢を継承し、この伝説の謎の秘宝を追っていた。トレジャー・ハントに多額の資金が必要なベンは、不本意ながら富と名声のために秘宝を探すイアン（ショーン・ビーン）と手を組む。暗号を解き進むうち秘宝の謎を解く鍵が「アメリカ合衆国独立宣言書」にあることを突き止めたベンだったが、イアンらの裏切りにより、命を狙われる。アメリカの魂ともいえる「独立宣言書」をイアンら金の亡者に冒涜されることを危ぶんだベンは、彼らよりも先に「独立宣言書」を入手し、貴重な歴史遺産と伝説の秘宝を探し出し、守ることを決意する。早速「独立宣言書」が収められたワシントンD.C.の国立公文書館を訪れると、そこには公文書管理者である聡明で美しいアビゲイル・チェイス博士（ダイアン・クルーガー）がいた。イアンとベンの激しい争奪戦に巻き込まれた博士ははじめ抵抗するが、やがて伝説の秘宝の謎解きに魅せられていく。
「独立宣言書」を手にしたベンとアビゲイル博士が英知を駆使して暗号解読に取りかかると、そこに謎の数字が浮かびあがってきた。これで、伝説の秘宝を見つけられる、と喜んだのも束の間、それはまだ、伝説の秘宝を解く鍵の入口でしかなかった。迫るイアンたちの追撃を振り切りながら、次々と現れる謎の暗号を解読していく二人。果たして伝説の秘宝にたどり着くことはできるのか・・・。
